Not while MAME is full screen.

If you ran MAME in a window, you'd have the mouse and keyboard, but MAME would lose focus as soon as you clicked on anything else. I'm not sure, but if your controller shows up as a joystick in windows, it might still work even when MAME doesn't have focus? Anyway, I'm guessing that's not what you're looking for.

Sweet setup, by the way. I run something kind of similar, except my "horizontal" screen is just a 20" CRT.
